Abstract

Osteoartritis merupakan penyakit sendi degeneratif yang ditandai dengan kerusakan bertahap kartilago artikular serta perubahan struktur sendi yang menimbulkan nyeri dan penurunan fungsi. Pada tahap awal, perubahan biologis berlangsung pada tingkat seluler sebelum kerusakan struktural teridentifikasi secara klinis. Penelitian ini bertujuan untuk mengkaji mekanisme patofisiologis yang mendasari disfungsi kondrosit pada fase awal osteoartritis. Metode yang digunakan adalah tinjauan sistematis dengan mengacu pada pedoman PRISMA 2020. Penelusuran literatur dilakukan melalui basis data PubMed, Scopus, dan Semantic Scholar dengan rentang publikasi tahun 2016–2026. Sebanyak 300 artikel ditemukan, kemudian diseleksi berdasarkan judul, abstrak, serta kriteria inklusi dan eksklusi yang telah ditetapkan. Dua artikel yang memenuhi kriteria dianalisis menggunakan Joanna Briggs Institute Critical Appraisal Tool. Hasil menunjukkan bahwa disfungsi kondrosit dipicu oleh aktivasi jalur inflamasi dan stres oksidatif. Selain itu, gangguan metabolisme seluler serta disfungsi mitokondria turut berperan dalam proses tersebut. Kondisi ini mengakibatkan ketidakseimbangan aktivitas anabolik dan katabolik matriks kartilago. Ketidakseimbangan tersebut berkontribusi terhadap peningkatan degradasi matriks dan percepatan progresi osteoartritis. Penelitian selanjutnya disarankan untuk mengembangkan pendekatan terapi yang menargetkan mekanisme molekuler kondrosit.
